What is Enalapril maleate and what is it used for
Enalapril maleate is the maleate‐salt form of the active substance enalapril, an angiotensin-converting-enzyme (ACE) inhibitor. It acts as a pro-drug: after oral administration it is converted into its active metabolite enalaprilat which inhibits ACE, thus reducing the formation of angiotensin II, leading to vasodilation and reduction of blood pressure.
The main therapeutic uses are treatment of hypertension (high blood pressure), symptomatic heart failure, and prevention of symptomatic heart failure in patients with left‐ventricular dysfunction.
How is Enalapril maleate produced
The synthesis of enalapril maleate involves multi‐step chemical reactions. One described route begins with the condensation of the benzyl ester of L-alanyl-L-proline with the ethyl ester of 3-benzoylacrylic acid, followed by reduction (e.g., hydrogenation with Raney nickel) to yield enalapril, which is then converted into its maleate salt form. The maleate salt (maleic acid combined with enalapril) improves stability and formulation properties.
In downstream manufacturing, formulation stability is an important concern: e.g., hot‐melt granulation techniques have been researched to improve chemical stability of enalapril maleate tablets.
How large is the global market for Enalapril maleate and where is it produced
The global market for enalapril maleate is estimated in the realm of around USD 1.6-1.7 billion in 2024. For example, one report states the market size was valued at USD 1.7 billion in 2024 and is forecast to grow to about USD 2.5 billion by 2035.
The active pharmaceutical ingredient (API) and finished dosage forms are produced globally, with major manufacturing centres in China (both API and formulations) and India, as well as production in the USA and Europe.
Specifically, Chinese suppliers dominate export of the API: one data source lists ~98 suppliers in China for enalapril maleate (HSN 2933) exporting globally, with three Chinese companies accounting for ~86% of China’s exports.
