What is Glibenclamide and What is It Used For
Glibenclamide (also known as glyburide) is a second‑generation sulfonylurea oral antidiabetic medication. It works by binding to ATP‑sensitive potassium channels (KATP) on pancreatic beta cells, causing depolarization, calcium influx and thereby stimulating the release of insulin. It is used in the treatment of type 2 diabetes mellitus when diet and exercise alone are insufficient to control blood glucose. It is available as a generic medicine and has been approved for use in many countries.
How is Glibenclamide Produced
The chemical synthesis of glibenclamide starts from substituted benzoic acid derivatives (for example 2‑methoxy‑5‑chlorobenzoic acid chloride), which is converted into an amide with 2‑phenylethylamine, then sulfonylation (e.g., chlorosulfonic acid) to form a sulfonamide intermediate, and then reaction with cyclohexylisocyanate to yield glibenclamide. Recent work has shown continuous‐flow methods can achieve higher yields (80‑85 %) for the semi‑telescoped steps within minutes. Commercial production of the active pharmaceutical ingredient (API) is typically carried out in GMP‑certified facilities, and suppliers of glibenclamide API can be found in India, China, Italy, France and elsewhere.
